چکیده

The deepest phylogenetic division in the universal tree (vertical line in Figure 1) is that separating bacteria from the clade comprising archaea and eukaryotes. The prokaryote–eukaryote split (horizontal line in Figure 1), originally delineated on the basis of differences between (eu)bacterial and eukaryotic cellular ultrastructure, is a phenetic dichotomy (see Doolittle, 1996, and references therein). How useful this dichotomy still is remains an active question: its answer will come from a full appreciation of molecular and cellular differences between

DNA REPLICATION AND SYNTHESIS OF DNABINDING PROTEINS IN THE CHLOROPLASTS OF A CALLUS CULTURE

Continuous labelling of callus with H-thymidine results in intermittent peaks of H-DNA per chloroplast, showing synchrony of division. The increase in H-DNA could be due to several replication rounds, and the drop to successive plastid divisions without intervening DNA synthesis. The level of DNA-binding proteins in the chloroplast parallels the peaks of plastidal DNA synthesis; such pro...

Diversity of DNA Replication in the Archaea

DNA replication is arguably the most fundamental biological process. On account of their shared evolutionary ancestry, the replication machinery found in archaea is similar to that found in eukaryotes. DNA replication is initiated at origins and is highly conserved in eukaryotes, but our limited understanding of archaea has uncovered a wide diversity of replication initiation mechanisms. Archae...

Cell Timer/Cell Clock

Like the biological clock in the body, replication of each cell type (even different cells of the same organism) follows a timing program. Abnormal function of this timer could be an alarm for a disease like cancer. DNA replication starts from a specific point on the chromosome that is called the origin of replication. In contrast to prokaryotes in which DNA replication starts from a single ...

Characterization of the replication initiator Orc1/Cdc6 from the Archaeon Picrophilus torridus.

Eukaryotic DNA replication is preceded by the assembly of prereplication complexes (pre-RCs) at or very near origins in G1 phase, which licenses origin firing in S phase. The archaeal DNA replication machinery broadly resembles the eukaryal apparatus, though simpler in form. The eukaryotic replication initiator origin recognition complex (ORC), which serially recruits Cdc6 and other pre-RC prot...

Varicella Zoster Virus (VZV) Origin-Dependent Plasmid Replication in the Presence of the Four Overlapping Cosmids Comprising the Complete Genome of VZV

The Varicella-Zoster Virus (VZV) genome contains both cis-acting and trans-acting elements, which are important in viral DNA replication. The cis-acting elements consist of two copies of oriS, and the trans-acting elements are those genes whose products are required for virus DNA replication. It has been shown that each of the seven genes required for ori-dependent DNA synthesis of Herpes Simpl...
